It's either/or in that situation. You don't bin and give the penalty try as well. If the ref hadn't been convinced the try would have been scored on the Manfredi one then he would have binned him and not given the try. Likewise on the Isa one if he had thought a Mamo would have scored then he would have given the penalty try and not binned Isa. Hope that makes sense

That's totally separate from foul play stuff. Can still be sent off for foul play and give away a penalty try (e.g Chris Hill this year)
